FOREWORD


Currently, in the industry of e-cigarettes, pouches, liquids and the like, one can categorise nicotine products whose composition includes nicotine, nicotine derivatives, nicotine salts or mixtures of acids with nicotine.

Due to the adverse health effects of smoking and the diffi-culty many smokers have in quitting completely, it would be beneficial to have an effective me-
thod of quitting smoking and, moreover, moving away from nicotine, which is highly addictive and toxic.

Nicotine acts on nicotinic cholinergic receptors, affecting most organ systems in the human body. It can play an important role in the development of cardiovascular disease, fertility disorders and contributes to cancer.

Nicotine is one of many alkaloids produced by tobacco, and methods have also been developed to synthesise this compound for commercial purposes.


NoNicTM is a brand-new alternative to traditional 
nicotine. We have patented this revolutionary substance all around the world, including the USA, UAE, Europe, Asia and other markets.


NoNicTM is a non-toxic, non-addictive and non-psychoactive substance, and has an effect just like nicotine. It can be used in both disposable e-cigarettes and nicotine pouches.


Alternative to traditional nicotine NoNicTM

There is widespread interest in chemical compounds related to nicotine, both for recreational and pharmaceutical applications.

One of these, a synthetic compound called 6-methyl nicotine, has been identified as a non natural alkaloid analog of nicotine. It has been targeted for its chemical structure, sensory properties and commercial availability.

Accordingly, chemical, pharmacological and toxicological evaluations of 6-methyl nicotine were carried out and compared with (S)-nicotine of pharmaceutical purity.

The 6-methyl-nicotine samples Analysed included both the free base and salt, as well as e-liquid formulations containing propylene glycol (PG) and vegetable glycerin (VG) for use in an electronic nicotine delivery system (ENDS).


ANALOG OF NICOTINE

Conclusions drawn from the results* indicate that 6-methyl nicotine has similar chemical properties to (S)-nicotine and behaves similarly in ENDS devices.


In silicon toxicological studies and historical pharmacological studies suggest somewhat comparable effects of the two compounds. 6-Methyl nicotine shows comparable toxicological behaviour to (S)-nicotine, with no mutagenic or geno-toxic effects and limited cytotoxicity.

Preliminary experiments therefore
suggest that the compound in question, 6-methyl nicotine, may be a suitable substitute for nicotine.


ANALOG OF NICOTINE


The aforementioned research was performed by Enthalpy Specialty Labs, Richmond, VA, USA,  and presented in the form of a Poster at the Tobacco Science Research Conference in August 2023. 

PHARMACEUTICAL RESEARCH SCIENTIFIC PUBLICATION


The available literature also includes an article describing an evaluation of the cytotoxicity of nicotine compared to 6-methylnicotine on human bronchial epithelial cells (BEAS-2B cells) in vitro [2]. The findings of this publication suggest that E-cigarettes containing 6-methyl nicotine may offer some advantages over conventional electronic cigarettes containing nicotine.

For example, considering one of a number of results from studies performed, where BEAS-2B cells were exposed to nicotine and 6-methyl nicotine at their maximum tolerated concentrations, one can see the conclusion drawn, which refers to the inhibition of expression of genes closley related to carcinogenesis only in the case of 6-methyl nicotine.

Moreover, previous literature reports [3], indicate that 0.3 mg of 6-methyl nicotine is equivalent to 1.0 mg of nicotine in terms of sensation and taste, although it has only a slight addictive effect. The very fact that the compound in question causes similar sensations used in a lower dose suggests that the classification of products with its content shows less toxicity, harm and addictive
potential.

UNDERSTANDING NONIC TM

As defined by the International Union of Pure and Applied Chemistry (IUPAC for short), which is dedicated to standardising symbols, nomenclature and standards of physical quantities used by chemists worldwide, alkaloids are defined as  a group of naturally occurring basic nitrogenous and organic compounds, mostly
heterocyclic, mostly of plant origin.

In addition, similar definitions are reproduced in the literature, for example, D.K. Semwal in his book “The essential guide to alkaloids” [1] states that alkaloids are, naturally occurring organic substances” containing, among other things, nitrogen and having an alkaline nature.

They are not necessarily of plant origin, as they can be synthesised by other living organisms, including some animals.

Nevertheless, their immanent feature is their origin in biosynthesis. Accordingly, if a compound is not produced by living organisms, it cannot be called an alkaloid.


As of today, there are no reports that the chemical compound known as 6-methylnicotine (systematically 2-methyl-5-[(2S)-1-methylpyrrolidin-2-yl] pyridine) occurs in nature in its pure form [1] Semwal, D. K., (2023).

On the contrary, in the absence of the possibility of extracting this compound from natural sources, various methods have been developed to obtain 6-substituted nicotine analogs by chemical synthesis, including 6-methyl nicotine, as evidenced by available scientific publications.


A strong argument that the compound in question can only be obtained by chemical synthesis is the characteristics of the pyridine ring, which makes it very difficult to undergo electrophilic substitution at the carbon atom, due to the significant reduction in electron density by the highly electro negative nitrogen atom.

The alkylation reaction, the attachment of a methyl (alkyl) group to the ring, is an example of such a substitution.

Therefore, the probability that such a reaction can occur in the environment decreases significantly and indicates the need for chemical synthesis methods.

In addition, there are numerous scientific reports referring to 6-methylnicotine as a synthetic compound, not derived from tobacco or nicotine.


According to the aforementioned arguments, the compound 6-methylnicotine cannot be conside-red an alkaloid.
